چکیده
Angiogenesis is the sprouting of new blood vessels from pre-existing ones. In physiological circumstances, angiogenesis is an essential process during development and wound repair. However, when dysregulated and persistent, angiogenesis can be a causal factor in a number of pathologies. Abnormal angiogenesis or endothelial cell function is involved in cancer, ischemic and inflammatory diseases. Therefore, angiogenesis is a tightly regulated process which includes endothelial cell proliferation, selective degradation of the basement membrane and the surrounding extracellular matrix, endothelial cell migration, and the formation of a tubular structure. Increased vascular permeability and cytoskeletal changes in endothelial cells lead to the disruption of endothelial cell junctions and therefore increased vascular permeability. A balance between proangiogenic and antiangiogenic factors controls the formation of new vessels. One of the most important proangiogenic factors is vascular endothelial growth factor (VEGF)1 .

Rho Kinase Inhibitors as a Novel Treatment for Glaucoma and Ocular Hypertension
In an elegant example of bench-to-bedside research, a hypothesis that cells in the outflow pathway actively regulate conventional outflow resistance was proposed in the 1990s and systematically pursued, exposing novel cellular and molecular mechanisms of intraocular pressure (IOP) regulation. The critical discovery that pharmacologic manipulation of the cytoskeleton of outflow pathway cells dec...
متن کاملRho-kinase mediates hyperglycemia-induced plasminogen activator inhibitor-1 expression in vascular endothelial cells.
BACKGROUND Elevated levels of plasminogen activator inhibitor-1 (PAI-1) are associated with myocardial infarction and stroke, especially in patients with diabetes. The induction of PAI-1 expression by hyperglycemia involves oxidative stress and protein kinase C (PKC). However, the mechanism by which hyperglycemia increases PAI-1 expression is unknown. METHODS AND RESULTS Compared with normogl...

Lipid nanocapsules (LNCs) represent a stable, biocompatible and worthwhile drug delivery system, demonstrating significant potential as gene/drug delivery platforms for cancer therapy. Imatinib, a potent tyrosine kinase inhibitor, has revolutionized the therapy of malignancies resulting from abnormal tyrosine kinase activity. However, its Clinical effectiveness in cancer treatment is h...
متن کاملRho kinase inhibitor HA-1077 prevents Rho-mediated myosin phosphatase inhibition in smooth muscle cells.
In smooth muscle, a Rho-regulated system of myosin phosphatase exists; however, it has yet to be established whether Rho kinase, one of the downstream effectors of Rho, mediates the regulation of myosin phosphatase activity in vivo. In the present study, we demonstrate in permeabilized vascular smooth muscle cells (SMCs) that the vasodilator 1-(5-isoquinolinesulfonyl)-homopiperazine (HA-1077), ...
متن کاملP130: The Role of Rho-Kinase (ROCK) in Microglia/Macrophage Polarization in Neuroinflammatory Diseases
Macrophage/microglia with heterogonous phenotype and function under physiological and pathological conditions are the main cell lineage involved in inducing immune responses in neuroinflammatory diseases which exhibit combined inflammatory and anti-inflammatory functions. An increase in the expression of iNOS triggers M1 phenotype that secrete high concentrations of inflammatory cytokines, whil...
